It varies by game. Generally, most titles keep mods in the Documents folder on the C: drive for the player.

It seems unlikely since it alters items in the game directory and removing high-resolution DLC gives you a bonus of 50 gigabytes of free storage.

For Fallout 4 and similar games, yes, you can. When you first launch Nexus, it prompts you about storing mods, etc. If you need to move your documents folder, right-click it, go to properties, and choose the new location. You can then set the documents folder on a separate drive with your games.

Nexus Mod Manager offers this functionality. I previously used it in Skyrim. My SSD handles the game, while my HDD stores all the mods.
