I increased my Rx 470 (Asus STRIX edition) with MSI afterburner and then enabled OC Mode in GPU Tweak 2. It will either boost performance further or remain unchanged, depending on the current settings.

Your clock speed is what you're using. It can be changed up or down. There isn't any "stacking" of overclocks; it's likely the ASUS overclocking tool will aim for a certain target speed when you enable OC mode, which could be higher or lower than your current afterburner setting. I believe it's wise not to run two programs competing for GPU control. It would be better to pick one program and remove the other.
