You're experiencing issues where the virtual box fails to detect your USB drive again. Despite installing an extension pack and adding your user to the vboxusers group, the problem persists between Ubuntu 20.04 (host) and MacOS Catalina (guest). Try restarting both systems or checking USB permissions.

This is likely the problem. VB macOS guest functionality is experimental and unsupported . There's no real guarantee anything will work. You can't even install guest additions... Try changing your USB settings, i.e. by enabling or disabling USB 2.0 support.

Yes, it does matter. Plugging in your USB before or after starting the guest affects how the setup proceeds.

Create a bootable USB drive. Choose the USB when prompted. Even after mounting, you should see its location in macOS.

It seems we overlooked Apple's unique formatting rules and their lack of standard USB support. I connected it to Windows, which reformatted it using Transmac, and it worked on macOS.
