Are you relying on the motherboard utility or flashback? If using a utility, they generally don’t need much attention anymore—they can locate the BIOS even within a large folder of files. If you’re using flashback, make sure you follow the instructions precisely for the flash drive. You’ll often see step-by-step guides on YouTube for most motherboards, e.g.:

I’m checking the instructions to confirm. It seems the USB should go into the flash slot, not the USB port itself.

These sections are marked for flashback, but you're not engaging with them.

The USB port labeled BIOS Flashback serves as a unique connection for automatic BIOS programming, bypassing the need to install a CPU directly into the socket. For this process, you might have to prepare your USB drive with FAT32 format and label the BIOS file appropriately, such as MSI.ROM or 7E06.BIN (where 7E06 represents your motherboard model). After that, activate the button on the board and allow 5 to 10 minutes for the BIOS to read from and write to your USB stick. To refresh the BIOS using the built-in utility, any standard USB connector on the back should suffice—this includes the BIOS Flashback port (compatible with USB 2.0 or 3.0, not high-speed ports). Simply insert the USB stick, select the BIOS file on it, and choose the file from your device. You may need to connect the stick before starting the BIOS update, as this ensures it recognizes your drive.
