ASUS provides a compatibility guide for this Thunderbolt card. You can view the list here: https://dlcdnets.asus.com/pub/ASUS/mb/Ad...200604.pdf This document notes compatibility with certain boards, which may include both WiFi and non-WiFi versions (such as the Z490-PLUS). The motherboard manual suggests it should work, though it isn't fully confirmed due to the missing board in the compatibility list.

You can adapt the 5-pin connector from your old motherboard to a 14-pin header by using a conversion board or adapter. This allows you to maintain compatibility while upgrading your system.

Thank you for bringing this up. It seems I misunderstood the product names. The motherboard OP supports "ThunderboltEX 3" (with a 5-pin connection) rather than "THUNDERBOLTEX 3-TR" (which uses a 14-pin connector). I didn’t realize the WiFi and non-WiFi boards differ in this regard.
