to be honest, i'm pretty sure its possible, but i doubt its a good idea. reads/writes (writes especially) can trash a USB drive rather quickly.
sidenote: i tried mineOS, and i'm not sure what to think of it... from one side it looks amazing, everything is very simple and easy to use. from the other side, it could be SO much more than just a dumbed down webmin interface. also, why the hell would you want a 20-person server on a portable media?
Running a 20-person server from your home PC and connection can quickly become problematic...
If it's only for a small group, it's okay—but keep it out of reach. For better performance, consider renting a modest server. Avoid using MC hosting services; they're unreliable. Companies like Online.net and OVH offer excellent Minecraft server packages at prices even MC hosts can't match. Side note: Online.net is French, and their English site doesn’t look very polished, which makes me a little doubtful about their quality.
