The math isn't too complex. One DDC would handle the setup. I'm using two MCP35X pumps in my custom loop. During filling and draining, I use a single pump to move a lot of coolant while keeping others off. Before my 1080 upgrade, I had two GTX 970s and blocks in the loop. In short, I have 2 360 rads, a 240 CPU block, a motherboard VRM block, two GPU blocks, a double-wide case with tubing, and lots of compression fittings. Coolant flows freely in the reservoir. I chose two pumps to reduce noise, RPM, heat, power draw, and add backup. Also, you said about one radiator—does that match what you have? If yes, is it a large external model?
