You're experiencing issues with your HDMI connection while the DP port works, but the DP port doesn't function at all. This is not uncommon when using Intel HD Graphics with certain setups. The problem might relate to cable quality, driver issues, or driver conflicts. Testing with a dedicated GPU and two monitors confirms the issue is specific to your main display setup.

The DP cable is DP on both sides, but HDMI requires special support from the source port.

With an HDMI to DP adapter, the connection functioned for roughly a year. The same cable and adapter also worked with the dedicated graphics card.

Same equipment used in identical configuration? Yes, the same hardware connected through the same method. The article discusses how some iGPUs provide DP output while others deliver DP++. Most discrete GPUs support DP++.

Initially assume the cable is the issue if the problem persists.

The identical cables function correctly with the dedicated graphics card, which is unusual given its stable performance over nearly a year. Could anyone confirm whether it's possible to turn off the DP port solely within the Asus BIOS settings?

It appears this feature is referred to as multi-display. Turning it off prevents running multiple monitors simultaneously from the CPU and GPU.
