I would put the backplate onto the motherboard and then install the motherboard itself. Once that's done, I'd arrange everything in the most convenient sequence for your specific situation. It really doesn't matter which way is easiest since there are too many factors involved. The decision mostly comes down to your judgment.

Personally, I install RAM first, then if it was the old HSF (heat-sink - fan) coolers I'd mount it then. When I did my Cryorig A40 I mounted the radiator and installed the motherboard and then attached the waterblock/pump. RAM can be installed at any time as there will be enough space with your AIO cooling. If you need a bracket/backplate though, remember to install that before putting in the motherboard.
