If sold as a complete unit, I’d leave out the cooler for shipping purposes, especially if it’s not standard and stands out. For single shipments, placing the Mobo in an anti-static bag with bubble wrap should suffice, or better yet, use the original box if available. Regarding the CPU, simply place it in its plastic tray and then wrap that in bubble wrap.

The anti-static bag matters whether it blocks static or shields against static electricity. As for the CPU, it should work just fine in the plastic tray even without the bag.

I’m not sure about the differences between the various bags, but an anti-static bag seems sufficient since manufacturers include them with the boards. If you still have the original bag from your motherboard, that would be ideal.
