Sure, you can copy the entire DVD content to a USB drive and use it as a bootable drive. Just make sure the copy is clean beforehand.

The USB partitions must be configured as bootable, or you may utilize Rufus.

It's important to prepare the USB flash drive so it boots up. Microsoft offers a useful tool that handles this automatically. Simply launch it, choose your ISO file, select the option to install on a USB drive, and it will manage everything for you. You can find it at: https://wudt.codeplex.com/

Yes, you can easily move the .ISO file to a thumb drive. Whether NTFS or FAT32 works doesn't matter. The main limitation is that the USB only supports UEFI systems. For better compatibility, it's best to make a bootable USB. I suggest using RUFUS because it's quick and simple. Other recommended tools have also been shared.
