When you enable XMP in your Asus UEFI, you’ll need to select "enable all core enhancement." The setting for "Asus multicore enhancement" should be set to auto by default. For overclocking with sync across all cores on an i5 6600k, adjust these options accordingly.
If you're pushing your CPU beyond overclocking limits, those adjustments won't make a difference. They force an unoverclocked processor to operate with more than one core at full turbo speed. This is the first time I've noticed this option. It seems the multicore boost can be enabled automatically? When you're exceeding the boost frequency, your core ratios might negate any benefits from this setting?
