You should use slots 2 through 8 for dual-channel support. This configuration allows both channels to be active simultaneously.
I don't suggest using three RAM sticks in that configuration. Instead, place the 4 and 4 in slots 1 and 3 or 2 and 4 as recommended by your motherboard manual. Put the 8GB in one of the remaining slots, ideally next to the first slot if you're only using one stick. The arrangement could look like (4GB - 8GB - 4GB - Empty) or (Empty - 4GB - 8GB - 4GB) depending on slot suggestions. This setup gives two 4GB sticks in one channel and a single 8GB stick in the other.
Consult the motherboard's guide, it will indicate which ports to use for 1, 2, 3 or 4 modules. If you lack a manual, look up your board's name on the web, especially the maker's site, where you'll usually find the documentation.
