The BIOS update process depends on your CPU compatibility. Since the X9DRE-TF+ needs at least BIOS 3.0 for v2 CPUs, you can install the update if your processor meets that requirement. You should be able to apply the BIOS upgrade without installing any CPU, but ensure your system meets the minimum BIOS version first. If not, you may need a compatible CPU to proceed.
You likely can install a BIOS update via IPMI, which doesn't need a functional CPU, though I haven't managed to get it working reliably on the Supermicro boards you've tested. Results might differ.
