Most users only found the TB4 card from Asrock, but some shared an adapter cable from Moddiy (link provided) that connects a 14-pin header to a 5-pin connector. I'm curious if it works with that adapter. Regarding BIOS, you might want to look into Memory Context Restore settings to speed up boot times. It depends on several factors. The adapter could function, the board might support the ASUS card, and the ASUS card could work with the board—though the Thunderbolt signal might need version 5. There’s no guarantee. Why would you require Thunderbolt 5? Desktop Thunderbolt behaves differently from laptop versions. What scenario matters most? Regarding boot times, it usually takes a while. It’s not your typical desktop; it’s more like a workstation with server-level components, so expect longer startup. The more RAM you have, the better the performance will be.

I mainly wanted TB connectivity and chose TB5 over TB4, so I figured someone tried that. In extreme cases, I might consider an Asrock TB4 AIC. If boot times are normal, it's fine. I've spent a lot of time on mobile platforms (NUCs), and many BIOS settings were missing. I'm looking into the latest BIOS updates—there are plenty of new options and I'd like to understand what they offer.
