If the BIOS allows PCIe splitting on the initial x16 port, and you aren’t planning to use a graphics card, it likely won’t function in the subsequent x16 port.

This might assist you with the discussion on AnandTech. You'll need a PCIe Bifurcation to accommodate that card. Certain premium cards come with their own chipset that handles this instead of relying on the standard motherboard chipset.
