Sure thing. The initial setup connects via PCIe 3.0/4.0 x4 to the CPU, and the second slot uses PCIe 3.0 x4 through the chipset. Both operate together at maximum speed.

The second position will operate only at 3.0 speeds, yet all drives will function because 4.0 remains backward compatible. Using a faster drive would be pointless as you wouldn't benefit from the higher speed. It would still work adequately.
