I set up a fresh boot choice, stored it, closed the BIOS, and after restarting, the boot option disappeared.

You needed to restore the boot option after saving. Initially, the feature disappeared when you restarted and saved. The solution involved transferring the SSD to your desktop PC, assigning it a letter in Windows Explorer, and then reconnecting it so you could boot from the USB drive again.
