Use the port near the BIOS flash button or another available port, but I favor the one next to the flash button. Please update the BIOS from the BIOS setup tool.

Yeah, what SavageNeo said. I prefer using the black USB ports by the PS2 connection and a USB stick that has LED activity light on it so I can see what's it doing. Be sure to extract the files from the ZIP file you download and put them on the USB stick. Don't just put the ZIP file on the USB stick. YOU MUST RENAME THE ROM FILE TO "MSI.ROM" If your computer boots just do it from in the BIOS. You can do it from dragon center in WIndows as well.' Edited April 8, 2020 by Paroxy Corrections.

I don’t own a USB drive with built-in lights. The Kingston 32GB Data Traveller you mentioned is likely a portable storage device, not a USB stick. Updates for such devices typically take a few hours to a couple of days depending on the complexity.

I only connect to the BIOS by pressing delete during startup.

Prepare the USB as FAT32 or exFAT. Load the extracted files from the ZIP into the drive, then reboot. Enter BIOS, click M-Flash, and proceed with the steps. It should be simple and take about 10 minutes or less.
