I think this might be a simple question, but I wanted to check if keeping the software open is necessary for the overclock to work properly.

Why not just use WattMan instead. THAT'S always running.
Here's a test. Close out your OC program and do a stress test while monitoring frequencies with something like GPUz. Does it match the OC profile you set?
