You should keep the USB drive handy during initial PC setup. Installing Windows 10 from it doesn’t require it to be present every time. Purchasing a key afterward won’t affect the OS if you don’t install the official Microsoft package.

After installing the OS on internal storage, you can detach the thumb drive without needing it attached. Windows will be ready whenever you want. No key is required during the first setup.

Initially, once you set up Windows 10 on a machine using a boot drive, you can later remove the drive and enjoy your fresh Windows installation. Also, it seems you haven’t activated Windows yet. If you purchased a key, you can use it to activate it afterward. Let me know if you need more details!
