You have two 2GB slots and plan to install 2x8GB drives on your iMac. It should work if the slots are compatible with the new drives.
No one can ensure compatibility between mixed ram modules. The safest choice is to purchase a kit in the desired size that has proven to work, and then replace your original ram. (or retain it for testing when issues occur)
