I recently reset my X99-E back to the default (F5) configuration to reinstall Windows. My main concern was whether I have to redo my overclocking or if it should be saved on the motherboard. I wasn't sure if changing the settings would erase the Profile I had set earlier.

Yes, you must redo all the overclocking unless you saved it as a profile. If you save it as a profile, you need to activate that profile so the system can apply the changes. Otherwise, at this moment the system is entirely factory non-OC with CPU, memory, and such frequencies/Voltages.

No, the F5 default restores the existing BIOS configurations, yet it doesn't erase the BIOS and OC Profiles as would occur during a BIOS update.
