Hi. The ASUS X99 e WS motherboard includes two 8-pin and one 6-pin connectors as shown in the diagram. Should I fill all these connectors? Since your PSU comes with an 8-pin cable, using four 4-pin connectors would work well to accommodate the dual slots. Regarding the PSU connections, you’ll need to determine where the 8 pins from the PSU go on the motherboard.

Link the CPU power plug marked "CPU/PCIe." The other end attaches to one of the eight pin CPU power connectors—you don’t have to use both. The additional one is only needed for extra power during overclocking. The same applies to the loose six pin power connector; it’s used just when several cards are connected, helping manage the 75W from the PCIe slots.

They are meant for practical use. The six-pin power connector won't be necessary.

I noted the power consumption from the ports. The PCIe six-pin adapter is included to help reduce the demand on the 24-pin port.

Additionally, it's not focused on SLI. The goal is to run several graphics cards at the same time. You don't have to rely on SLI/Crossfire to achieve this.
