Ensure your X16 card is plugged into the highest available X16 port, typically the one directly connected to the CPU. The next port is often reserved for X4 and should only be used for display output, not high bandwidth tasks. For M.2 slots, you can use any slot if routed through the chipset; however, AMD CPUs provide dedicated lanes for M.2 from the CPU, usually the top slot, so it won’t interfere with your X16 card.

Additionally, this hyper M.2 card needs a motherboard with PCIe bifurcation support, a feature available on some ASUS models but not all manufacturers offer it on ATX boards. This particular board appears to handle it.
