I intend to purchase a Z270 board and check if I can achieve overclocking with an Intel i5 6500. I aim to upgrade to a 7th generation processor within six months, so first I need to confirm if the Z270 supports overclocking.

Only adjusting the bus rate can lock the chips core clock. It's unclear what the maximum bus rate is before the system becomes unstable.

You're able to adjust the bus speed. In practice, you won't really feel any change, so it's best to wait for the update.
